In the Lab
Trying to do transformations
Made own competent cells and plates

Changed the protocol, now it works

Modeled what parts we need for the project
Identified parts we don’t have, those we do – see presentation slides (on annie’s computer)
UT Austin and Penn State, have what we need, gave them our address and haven’t heard since then
James Brown came to visit, we rearranged our diagram to fit the structure suggested by iGEM
Consider parts synthesis for project?
Making parts is more difficult than making devices
Realistically could make one or two parts with the time we have
Pull a piece of magnetic bacteria and put it into e.coli - would be a new part
Magnetotacic Bacteria
Ana brought some chemicals, Azeem and Jason have some as well
All other chems have been ordered, should be here by Monday
Can also get another batch of magnetotactic bacteria through prof. Palmore when we want it
Looking at coat protein of magnetosome
Attach a protein to the known coat gene then lyse them and isolate particles w/ the proteins attached
Put into eukaryotic cell, see if it recognizes it
If does, can then signal cell to do something else
Right now at the stage of making the media
Can make the mineral solution, then mix with a vitamin solution and another solution
Make it more iGEM?
KO of coat protein and then put the part on the plasmid
How do you then regulate this gene? Does it even matter?
For next week, we should read more about the bacteria
Ongoing library of articles for the wiki?
Manipulate the plasmid in E.coli and then put it into the magnetotactic bacteria
Have an easier goal and peptide, that are useful to something else as well as a cancer specific protein
Useful to repeat experiment with luciferase if we can afford it
Is there a plasmid available? can we get it now so that we aren’t waiting?
Journal Club
On Tuesday, Azeem will present and then Peter the week after
Ana can’t make it
What happened to binder of hard copies of the papers we read? Victoria?
We will also discuss the paper Hayato sent out on magneto. bateria
Quick Things
We do need bio-hazard training, keep an eye on email
People will be coming around on Mon or Tues to put in bins, etc.
They will set up the waste unit, a glass disposal box comes from the custodian
Can get red biohazard bins from ana’s lab
