Also can work up the solid phase hsl bomb experiments. We will start these using minimal media A (unbuffered) at 37C as in basu et al., nature, 2005. they solidify their media with 1.5% agarose. We can use seakem LE agarose (the cheap stuff) for this purpose to begin with. The plates should be Amp/Kan because [http://rosalind.csail.mit.edu/r/parts/partsdb/view.cgi?part_id=5414 I15030] is in psb3k3 and all of the reporters are in psb1a2. also, we should probably use S-gal in the plates. *Rachel Building R0065.B0034.LacZalpha We need to submit the correct LacZalpha sequence into the registry, and give it a biobricked name. Then we need to submit R0065.B0034 as a part, and R0065.B0034.LacZalpha as a part. We will hold off on making F2620.E0430, because that is already constructed at MIT ([http://rosalind.csail.mit.edu/r/parts/partsdb/view.cgi?part_id=5121 I13273]) and we will be getting it in a week or two. Moreover, I6042 will function just like I13273 if it is provided with a source of LuxR. *Laura Prep out pSB1A2.[http://rosalind.csail.mit.edu/r/parts/partsdb/view.cgi?part_id=4471 I6042] (MIT 4.04) and cotransform into omnimax cells with psb3k3. [http://rosalind.csail.mit.edu/r/parts/partsdb/view.cgi?part_id=5414 I15030]. this will be a yfp tester construct for the hsl bomb. Also test pSB1A2.R0065.B0034.LacZ made by JT last year as a reporter for the edge detector. This construct has 6 mutations, so we need to see if it still produces a functional LacZ protein *Dan Dan is currently deriving a mathematical model to analyze the light wires design.
