the meeting was also quite productive:
- we got kris up to date and he is about to model everything one could possible model regarding this module - well, hopefully =)
- giorgia presented her findings. 2 solutions remain:
- the basic mechanism with repressor and activator separated (not so sexy, the two mechanisms have to be adapted to each other)
- lambda-phage system (new: only one regulator, CI, has to be produced to achieve both effects, repression and activation and it is a standard and well known part)
- herve's solutions - unfortunately, he was away, so we will catch up on his findings soon
- discussion of how to incorporate the lambda-system with the zinc-fingers. open questions are
- where to insert the zinc-fingers: in the promotor (entangled systems) or rather between start and RBS or RBS and gene (separated systems). obviously, the latter seems much more feasible with lower risk.
- whether or not we should cut out the 3rd and last binding region. this could affect the overall performance. probably, we should test both approaches, original and cut, in parallel.
