Also can work up the solid phase hsl bomb experiments.  We will start these using minimal media A (unbuffered) at 37C as in basu et al., nature, 2005.  they solidify their media with 1.5% agarose.  We can use seakem LE agarose (the cheap stuff) for this purpose to begin with.

Sequence verified I15030 in pSB3K3.  Also, double transformations are complete and successful (I15030 with I6042 and I15030 with R0065.B0034.Lacz [made by Jeff]).  Testing for functionality of the HSL bomb today.  Also, I will make a freezer stock of circuit 1 (HSL bomb, I15030) in pSB3K3.  Here we go...

Building R0065.B0034.LacZalpha We need to submit the correct LacZalpha sequence into the registry, and give it a biobricked name. Then we need to submit R0065.B0034 as a part, and R0065.B0034.LacZalpha as a part.

We will hold off on making F2620.E0430, because that is already constructed at MIT ([http://rosalind.csail.mit.edu/r/parts/partsdb/view.cgi?part_id=5121 I13273]) and we will be getting it in a week or two. Moreover, I6042 will function just like I13273 if it is provided with a source of LuxR.
