Can work up (and show rachel, laura and jeff) how to do liquid phase LacZ assays. Also can work up the solid phase hsl bomb experiments. We will start these using minimal media A (unbuffered) at 37C as in basu et al., nature, 2005. they solidify their media with 1.5% agarose. We can use seakem LE agarose (the cheap stuff) for this purpose to begin with.

Building R0065.B0034.LacZalpha We need to submit the correct LacZalpha sequence into the registry, and give it a biobricked name. Then we need to submit R0065.B0034 as a part, and R0065.B0034.LacZalpha as a part.
We will hold off on making F2620.E0430, because that is already constructed at MIT ([http://rosalind.csail.mit.edu/r/parts/partsdb/view.cgi?part_id=5121 I13273]) and we will be getting it in a week or two. Moreover, I6042 will function just like I13273 if it is provided with a source of LuxR.

Prep out pSB1A2.[http://rosalind.csail.mit.edu/r/parts/partsdb/view.cgi?part_id=4471 I6042] (MIT 4.04) and cotransform into omnimax cells with psb3k3. [http://rosalind.csail.mit.edu/r/parts/partsdb/view.cgi?part_id=5414 I15030]. this will be a yfp tester construct for the hsl bomb. Also test pSB1A2.R0065.B0034.LacZ made by JT last year as a reporter for the edge detector. This construct has 6 mutations, so we need to see if it still produces a functional LacZ protein
