Plasmid Prep

Using overnight culture of plasmid I13522 (pTet-GFP), do plasmid prep on all 8 of the 5 ml cultures.

Combine the eluted plasmid from all the preps into one tube and measure the yield as follows:

1. turn on UV spec and blank with H2O
2. measure A260 of 4 ul plasmid + 76 ul H2O
3. this measurement gives the concentration in ug/ul, multiply by 1000 to give ng/ul
