The Lac stochastic trigger device serves as the stochastic random element that can switch cells into the ON state. LacI (C0012) is continually made via the constitutive promoter (R1075). LacI negatively regulates the LacI promoter (R0010), preventing any TIPS from going out to the switch input. We will manually input a certain concentration of IPTG into the system media. IPTG represses the LacI repressor, thereby positively regulating the LacI promoter. Based on the concentration of IPTG, we can tune the noise level of this procedure such that activation of the promoter occurs with low probability. Those cells that turn on the promoter become the sender cells in our system and begin attracting neighbors.
