Plasmid prep was done by Brad and Eric and I (kelly) jumped in to help with the A260 readings.  We obtained 71 ng/uL.

Do plasmid prep on pellets in three tubes labelled "GFP TE" in bottom of left fridge. They are from overnight culture of plasmid I13522 (pTet-GFP).

Combine the eluted plasmid from all the preps into one tube and measure the yield as follows:

  1. turn on UV spec and blank with H2O
  2. measure A260 of 4 ul plasmid + 76 ul H2O
  3. this measurement gives the concentration in ug/ul, multiply by 1000 to give ng/ul

Plasmid prep was done by Brad and Eric (Followed plasmid prep protocol), and I (Kelly) jumped in to help with the A260 readings. We obtained 71 ng/uL.
