Plasmid Prep

Using pellets from overnight culture of plasmid I13522 (pTet-GFP), do plasmid prep on pellets in three tubes labelled "GFP TE" in bottom of left fridge.

Combine the eluted plasmid from all the preps into one tube and measure the yield as follows:

1. turn on UV spec and blank with H2O
2. measure A260 of 4 ul plasmid + 76 ul H2O
3. this measurement gives the concentration in ug/ul, multiply by 1000 to give ng/ul
